چکیده

Morphological changes in a dry phospholipid film on a solid substrate were studied below and above the main transition temperature, between the gel and liquidcrystalline phases by phase-contrast microscopy and AFM. A Phospholipid film in the liquid-crystalline phase exhibits flat, smooth layering, whereas that in the gel phase shows rough, random layering. These film morphologies are discussed in relation to the ability to form giant vesicles through the natural swelling method.

Hydration process of multi-stacked phospholipid bilayers to form giant vesicles

– The hydration process of multi-stacked phospholipid bilayers to form giant vesicles was investigated by time-resolved small angle X-ray scattering. The hydration of lipids in the liquid-crystalline phase was found to proceed in two stages. In the early stage, until about 100 s after hydration, lipid bilayers on a solid substrate swell about 12 Å and reach a quasi-stable state. In the late sta...

Incorporation of amphotericin B into large unilamellar vesicles composed of phosphatidylcholine and phosphatidylglycerol.

The spontaneous incorporation of the polyene antibiotic amphotericin B from a micellar solution into phospholipid vesicles was examined as a function of the lipid composition of the vesicles and their physical state. Virtually no insertion of the antibiotic into egg phosphatidylcholine vesicles was observed even when cholesterol was also present in the bilayer. In contrast, rapid incorporation ...

The mechanism of vesicle formation.

Phospholipid molecules exhibit amphiphilic properties and therefore they aggregate either in their crystalline state or in polar solvents into ordered structures with typical lyotropic liquid crystalline symmetries (de Gennes, 1974). At high lipid concentrations in water these are predominantly lamellar phases (Luzzati, 1968), while in aqueous solutions phospholipid molecules normally form self...

Interaction of apolipoprotein A-I in three different conformations with palmitoyl oleoyl phosphatidylcholine vesicles.

Interactions of apolipoprotein A-I (apoA-I) with cell membranes appear to be important in the initial steps of reverse cholesterol transport. The objective of this work was to examine the effect of three distinct conformations of apoA-I (lipid-free and in 78 A or 96 A reconstituted high density lipoproteins, rHDL) on its ability to bind to, and abstract lipids from, palmitoyl oleoyl phosphatidy...

Visualization and analysis of apolipoprotein A-I interaction with binary phospholipid bilayers.

Apolipoprotein A-I (apoA-I) interaction with specific cell lipid domains was suggested to trigger cholesterol and phospholipid efflux. We analyzed here apoA-I interaction with dimyristoylphosphatidylcholine/distearoylphosphatidylcholine (DMPC/DSPC) bilayers at a temperature showing phase coexistence. Solid and liquid-crystalline domains were visualized by two-photon fluorescence microscopy on g...
